Qualitative Analysis

Qualitative research is a method of inquiry employed in many different academic disciplines, traditionally in the social sciences, but also in market research and further contexts.[1] Qualitative researchers aim to gather an in-depth understanding of human behavior and the reasons that govern such behavior. The qualitative method investigates the why and how of decision making, not just what, where, when. Hence, smaller but focused samples are more often needed, rather than large samples.

Qualitative methods produce information only on the particular cases studied, and any more general conclusions are only propositions (informed assertions). Quantitative methods can be used to seek empirical support for such research hypotheses.

Quantitative Analysis

Quantitative analysis, in chemistry, is the determination of the absolute or relative abundance (often expressed as a concentration) of one, several or all particular substance(s) present in a sample.

Once the presence of certain substance(s) in a sample is known, the study of their absolute or relative abundance can help in determining specific properties. Knowing the composition of a sample is very important and several ways have been developed to make it possible, like gravimetric and volumetric analysis. Gravimetric analysis yields more accurate data about the composition of a sample than volumetric analysis does, but the first one takes more time to perform in the laboratory. Volumetric analysis in the other side doesn’t take that much time and the results that we obtain are in the most cases satisfactory. Volumetric analysis can be simply a titration based in a neutralization reaction but it can also be a precipitation or a complex[disambiguation needed] forming reaction as well as a titration based in an redox reaction. However, each method in quantitative analysis has a general specification, in neutralization reactions, for example, the reaction that occurs is between an acid and a base, which yields a salt and water, hence the name neutralization. In the precipitation reactions the standard solution is in the most cases silver nitrate which is used as a reagent to react with the ions present in the sample and to form a high insoluble precipitate. Precipitation methods are often called simply as Argentometry. In the two other methods the situation is the same. Complex forming titration is a reaction that occurs between metal ions and a standard solution that is in the most cases EDTA (ethylene diamine tetra acetic acid). In the redox titration that reaction is carried out between an oxidizing agent and a reduction agent.

For example, quantitative analysis performed by mass spectrometry on biological samples can determine, by the relative abundance ratio of specific proteins, indications of certain diseases, like cancer.

Quantitative vs. Qualitative Analysis

The general expression Qualitative Analysis [...] refers to analyses in which substances are identified or classified on the basis of their chemical or physical properties, such as chemical reactivity, solubility, molecular weight, melting point, radiative properties (emission, absorption), mass spectra, nuclear half-life, etc. Quantitative Analysis refers to analyses in which the amount or concentration of an analyte may be determined (estimated) and expressed as a numerical value in appropriate units. Qualitative Analysis may take place without Quantitative Analysis, but Quantitative Analysis requires the identification (qualification) of the analyte for which numerical estimates are given.

Internal Standard

An internal standard in analytical chemistry is a chemical substance that is added in a constant amount to samples, the blank and calibration standards in a chemical analysis. This substance can then be used for calibration by plotting the ratio of the analyte signal to the internal standard signal as a function of the analyte concentration of the standards. This is done to correct for the loss of analyte during sample preparation or sample inlet. The internal standard is a compound that is very similar, but not identical to the chemical species of interest in the samples, as the effects of sample preparation should, relative to the amount of each species, be the same for the signal from the internal standard as for the signal(s) from the species of interest in the ideal case. Adding known quantities of analyte(s) of interest is a distinct technique called standard addition, which is performed to correct for matrix effects.

This ratio for the samples is then used to obtain their analyte concentrations from a calibration curve. The internal standard used needs to provide a signal that is similar to the analyte signal in most ways but sufficiently different so that the two signals are readily distinguishable by the instrument. For example deuterated chlorobenzene (C6D5Cl) is an internal standard used in the analysis of volatiles on GC-MS because it is similar to Chlorobenzene but does not occur naturally. Nor-Leucine is also a popular internal standard for the analysis of amino acids via GC-MS.

In NMR spectroscopy of the nuclei 1H, 13C and 29Si, frequencies depend on the magnetic field, which is not the same across all experiments. Therefore, frequencies are reported as relative differences to the internal standard tetramethylsilane. This relative difference to TMS is called chemical shift, and measured in parts per million.

In practice, since the difference between the deuterium signals of the solvent and TMS are known, no TMS may need to be added. By specifying the lock solvent to be used, modern spectrometers are able to correctly reference the sample; in effect, the deuterated solvent serves as the internal standard.

According to the UNODC and the NIDA, TLC, as a method, is NOT accurate for the determination of relative or absolute LEVELS of cannabinoids in cannabis hemp or marijuana! To gain a more quantitative determination of cannabinoids in a sample, an internal standard will need to be analyzed with the sample in question. If an internal standard is used, a more accurate quantitative analysis can be made using thin layer chromatography.

To get TRUE quantitative analysis, a Gas Chromatograph (G.C) or High Pressure Liquid Chromatograph (H.P.L.C) should be used to quantify THC, CBD, CBN levels in a medical marijuana sample.

Gas v. Liquid Chromatography

Gas chromatography and liquid chromatography differ in one way that is important to the analysis of cannabinoidsGas chromatography uses heat and liquid chromatography does not.  Heat allows the sample of marijuana to go through a chemical process called decarboxylation, which is a necessary step to determine the exact amount of CBD and THC that can be absorbed into the body.  The body will not absorb marijuana that has not been decarboxylated, which is why prior to a patients use marijuana is always exposed to heat.  Marijuana is decarboxylated either through smoking, vaporizing, or cooking butter. Analyzing medical marijuana without decarboxylation, like through liquid chromatography, allows only for the measurement of the maximum THC and CBD possible in a sample.  This produces high numbers that are not useful to patients because they will need to decarboxylze marijuana in order to feel its effects.

This is why Gas Chromatography has been the method of choice for the quantification of cannabinoids by both the United Nations Office on Drugs and Crime (UNODC) and the National Institute on Drug Abuse (NIDA).

Abstract:

A GC–MS method was performed to determine the total 9-THC content in both drug- and fiber-type cannabis seeds. Drug-type seeds were found to contain much higher levels of 9-THC (35.6–124 g/g) than fiber (hemp) seeds (0–12 g/g). The majority of 9-THC was found to be located on the surface of the seeds. Approximately 90% of the total 9-THC was removed by a simple, quick wash with chloroform. Washed drug-type seeds contained less than 10 g/g. Separation of the seeds into the kernel and testa showed that the bulk of 9-THC is located in the testa, mainly on the outside. The kernels of drug- and fiber-type cannabis seeds contained less than 2 and 0.5 g 9-THC/g seeds, respectively. Fluctuations in the 9-THC content of different replicates of the same type of seeds could be the result of the degree of contamination on the outside of the seeds.

1. Introduction
1.1 Background

Cannabis products are the most widely trafficked drugs worldwide, accounting for 65 per cent of all global seizure cases (1.65 million cases) in 2006. 5,200 metric tons of herb and 1,000 metric tons of resin were seized in 2006. Practically all countries in the world are affected by cannabis trafficking. Similarly, cannabis also remains the most widely used drug worldwide, with an estimated 166 million people having used cannabis in 2006, equivalent to some 4 percent of the global population aged 15-64.

At the same time, especially since the end of the last century, production methods have become increasingly sophisticated, resulting in the availability in illicit markets of a wide range of cannabis products with widely varying levels of the main psychoactive ingredient, delta-9-tetrahydrocannabinol (THC). Most recently, there has lso been a renewed debate about increasing THC content (frequently referred to as “potency”) in illicit cannabis products. All of this requires analytical data which are comparable between laboratories and over time.

However, most countries do not require by law the detailed analysis of the THC content of the different products, and where such analyses are carried out, they use a variety of approaches and experimental designs, reducing the comparability of results. For example, the conversion of natural constituents, such as tetrahydrocannabinolic acid (THCA), by both smoking and under certain analytical conditions into THC, and how this should be reflected in the analytical report, are issues which are not yet standardized worldwide.

On the technological side, the analysis of cannabis products is further complicated by the relatively restricted availability of pure or well defined reference material of THC and other cannabinoids.* The present manual is an updated and significantly revised version of the manual on “Recommended methods for testing cannabis” (ST/NAR/8), which was published in 1987. It has been prepared taking into account both developments in analytical technology and advances in the science of cannabis, and with a view to providing the analytical basis for an objective discussion about changes in THC content over time, and differences between regions and products.

 “…This is just a knock off of the cannanlyse thc testing kit but with out a lot of supplies that come with the cannalyse kit ,also even though TLC is a common way of testing elements the dye and extraction fluids must be correct and the directions must be exact that come with the kits. TLC was the method of choice for this study as it provides a means of rapidly and visually analysing a number of samples simultaneously. Unlike the comparison of two samples in order to assess whether they are the same (or different), when quantitative analysis is essential, assessment of origin is qualitative, since the sample under study and the reference samples are, in fact, different. There is thus little point in using more powerful techniques such as high-performance liquid chromatography as the quantitative information provided is likely to confuse the picture. The TLC system was designed to give information on the cannabivarins in addition to the more commonly studied cannabinoids. The principal cannabinoids observed using this system are given in table 2. Sample preparation and detection were as previously described [ 47] [ 48] . The solvent system used was a mixture of chloroform (ethanol-free) and l,l-dichloroethane (15:10) with simple ascending TLC on 10 x 20 cm silica gel pre-coated plates with a layer thickness of 0.25 mm (E. Merck, Darmstadt, G.F.R., Art. No. 5729) [ 46] . Each extract was analysed within one hour of preparation. Twenty different samples of herbal cannabis and cannabis resin from several different countries were subjected to hot-solvent extraction to simulate the illicit preparation of liquid cannabis. The resulting chromatograms were compared with those of the starting material..

The control in this test was ~17% THC by volume… You can tell the size of the dots for Connie Chung (A) and (B) are larger. You can draw you own conclusions!

Abstract
The use of Cannabis is illicit in numerous countries and the increasing consumption has led to a multiplication of scientific studies. New methods of planar chromatography such as Automated Multiple Development (AMD) and Optimum Performance Laminar Chromatography (OPLC) techniques can be used as a substitute for the traditional TLC for the identification and quantification of the Indian hemp components. Each method offers its own advantage: high resolution with neither diffusion nor spot stretching for AMD, speed, efficiency and the possibility of working in semi-preparative mode for OPLC.

I received a comment on my blog recently mentioning the comparison of Alpha Nova’s Cannalytics™ cannabinoid thin layer chromatography testing system and Montana Biotech’s Grow Buddy™ T.L.C. test kit. Thin layer chromatography is a method for testing different compounds using a solvent and glass plates coated with silica gel and a dye for visualizing the results. The method is OLD, and nobody has the patent on it! According to the UNODC, T.L.C is not used for quantification, but THC,CBD, CBN, qualification. The thin layer chromatography method must be used with a standard to be quantitative in any capacity.

UPDATE: STOP SB423 Repeal in Disguise Petition / Voter Referendum Signature Collection Drive May 28, 2011 UPDATE!


!!UPDATE STOP SB423, Medical Marijuana Repeal in Disguise, Referendum, Injunction, Petition Signature Collection, Gathering Drive, June 20th- 21st UPDATE!!

As most of you now know, the language for the referendum to stop SB 423, repeal in disguise, from going into effect must be approved by the Secretary of State, Montana Legislative Services, and the Attorney General’s Office. The Secretary of State had approved the language and sent the petition on to Legislative Services. As a result of its review, Legislative Services has returned the language to the MTCIA with their concerns, which was an expected and anticipated part of the process. Not only does their input allow us the opportunity to refine the referendum language, it also provides insight as to any pitfalls that could be encountered up the road if medical cannabis opponents want to make trouble for us (which they will) by attacking minutia in the referendum’s language.

So, at this time, the language is being tweaked by an attorney to address the concerns of Legislative Services. These tweaks will make the referendum language more bulletproof.

Once we respond and submit a refined version, the proposed petition will continue on to the next steps required before a final petition receives formal approval for signature-collecting.

Thanks to everyone for their eagerness, and indeed impatience, in wanting to get this petition on the streets and racking up the signatures on behalf of Montana patients, voters, businesses, and families.

TLC, UNODC (United Nations Office on Drug and Crime)

TLC was the method of choice for this study as it provides a means of rapidly and visually analysing a number of samples simultaneously. Unlike the comparison of two samples in order to assess whether they are the same (or different), when quantitative analysis is essential, assessment of origin is qualitative, since the sample under study and the reference samples are, in fact, different. There is thus little point in using more powerful techniques such as high-performance liquid chromatography as the quantitative information provided is likely to confuse the picture. The TLC system was designed to give information on the cannabivarins in addition to the more commonly studied cannabinoids. The principal cannabinoids observed using this system are given in table 2. Sample preparation and detection were as previously described [ 47] [ 48] . The solvent system used was a mixture of chloroform (ethanol-free) and l,l-dichloroethane (15:10) with simple ascending TLC on 10 x 20 cm silica gel pre-coated plates with a layer thickness of 0.25 mm (E. Merck, Darmstadt, G.F.R., Art. No. 5729) [ 46] . Each extract was analysed within one hour of preparation. Twenty different samples of herbal cannabis and cannabis resin from several different countries were subjected to hot-solvent extraction to simulate the illicit preparation of liquid cannabis. The resulting chromatograms were compared with those of the starting material..

The control in this test was ~17% THC by volume… You can tell the size of the dots for Connie Chung (A) and (B) are larger. You can draw you own conclusions!

Abstract
The use of Cannabis is illicit in numerous countries and the increasing consumption has led to a multiplication of scientific studies. New methods of planar chromatography such as Automated Multiple Development (AMD) and Optimum Performance Laminar Chromatography (OPLC) techniques can be used as a substitute for the traditional TLC for the identification and quantification of the Indian hemp components. Each method offers its own advantage: high resolution with neither diffusion nor spot stretching for AMD, speed, efficiency and the possibility of working in semi-preparative mode for OPLC.
