Grow Buddy Pro

#1 Collect/Select your samples for analysis.

Use the Grow Buddy Pro supplies to collect and categorize your samples. This can be hash oil, kief, or dried plant material.

#2 Assay your samples.

Use Grow Buddy Pro to quantify cannabinoids content in your samples. Know what percent THC, CBD your sample/s contain.

#3 Analyze the results!

Use JustTLC software, by Sweday, with the Grow Buddy Pro THC, CBD, CBN pre-loaded TLC plates to determine the percentage THC, CBD, CBN of your samples.

Grow Buddy Pro:

Grow Buddy Probasically a Grow Buddy, but has a THC, CBD, CBN sample in the kit to help quantify percentages using a FREE software download by Sweday, JustTLC $150 plus S/H, retail.

Grow Buddy Pro Quantitative Assay

Area Prep:

1.) Remove all items from the area not needed for your testing.

2.) Collect materials not included in the kit needed for testing.

a) Thermometer

b) A scale accurate to .1g

c) Timer, or some way to keep track of elapsed time

d) Some way to capture an image of the T.L.C. plates after being sprayed with the T.L.C. dye.

          1. Flat scanner
          2. Digital camera

e) Some way to heat your dried samples, like a stove or toaster oven.

Now you are ready to start some testing!!

WEAR GLOVES!!

Sample Prep:

1.) Make sure samples are DRY and crumbly. The greater the moisture content the less accurate this T.L.C. method is.

2.) Give the samples a designation and record on a sheet of paper. Weigh out the samples in 1 gram increments on a piece of aluminum foil shaped like a bowl. Be sure to take note of the foil’s weight, and the sample plus the foil’s weight.

EXAMPLE: Place the foil on the scale, record weight: 200mg. Tare the foils weight. Place the sample on the foil trying to accurately weigh 1 gram as close as possible. Record weight: 995.6mg. Add together and record the total 1195.5mg with the samples designation.

3.) Pre-heat oven to 120°C- 250°F. Heat samples on the aluminum foil for 15 min.

4.) Subtract the samples before heating weight from the after heating weight, record the difference on a piece of paper for later. This is your activated dry weight (ADW).

NOTE: If the sample has melted, or is unable to be removed from the foil, easily, let the aluminum cool to room temperature. Use a large pipette and add 1mL of the Extraction Solution to remove the material from the foil. Place this solution into a 1.5mL tube with the large pipette. This solution represents ~1000mg not 100mg (10x more concentrated! Not to mention, the sample is probably some type of a concentrate to begin with!).

5.) Weigh 100mg of the activated dry sample in the 1.5mL tubes and add 1mL of the Extraction Solution.

Grow Buddy Pro T.L.C. Plate Prep:

1.) Only handle Grow Buddy™ Pro T.L.C. plates with gloved hands.

2.) Load 2 microliters of the Extraction Solution from the sample 1.5mL tube per into one of the four sample wells, skipping the middle well, well #3 contains ~2micrograms THC, CBD, CBN, or ~10% cannabinoids by dried weight. Note which sample designation went where. Note if the sample was plant material, flowers, hashish, kief, BHO isolate, wax, shatter, etc.

Grow Buddy Pro T.L.C. Plate Developing Prep:

1.) Place the contents of the developing solution bottle into the jar/developing chamber.

2.) Place the chamber on a level surface, and in an area where it will not be disturbed for ~15-20 minutes.

3.) Place Grow Buddy Pro T.L.C. plate into the chamber, carefully. Place the lid on the developing chamber, carefully. Let the plate run for ~15-20 minutes, or until the developing solution is ¼” from the top of the T.L.C. plate.

4.) Remove from the chamber and let air dry before using the T.L.C dye.

Grow Buddy Pro T.L.C. Dye Prep:

This is a DYE and WILL stain!!

1.) As the T.L.C. plates are running in the developing chamber, prepare your T.L.C. dye.

2.) Add 1mL water to the T.L.C. dye 1.5mL tube. Shake vigorously. Let set.

3.) Add 4mL of water to the T.L.C. sprayer.

4.) Transfer the 1mL T.L.C. dye concentrate to the T.L.C. sprayer. Shake vigorously. Let set.

5.) Use the Grow Buddy Pro box as a spray chamber.

6.) One 5mL sprayer will dye 2 plates.

7.) Spray plates as you practiced.

8.) Let air dry. Okay, you are almost done.

Capturing an Image:

By this point you have chosen a way to image your plates so you can save and analyze the data at your leisure. Have the plate imaged at least 15 min after dying. If not, results are unreliable.

How this list came about-

“If the truth won’t do, then something is wrong!”

Those were the furious words of my grandfather to my Mother. I had walked in from joyfully stuffing my face with red raspberries in the garden, straight into “war zone”! My gentle grandfather in a fury, his hand raised! Mom was just beginning to shrink back away from him. They saw me and quickly sent me away. But it was too late, the scene and the words were seared into my 5-year-old brain. That was over 55 years ago, but I still remember it clearly. My grandfather was a minister, one very short step away from God in my 5 year old mind. It was one of those life changing moments. It is still rare for me to tell a lie. I never found out what my Mother’s lie was.

As I child, I suffered a traumatic head injury. Another child tried to murder me with a hammer. I was left with frequent migraines. At 19, like many rebellious teens, I tried cannabis. It took about a year for me to make the connection between using cannabis and the absence of my normally frequent migraines. I have used cannabis ever since.

I am an avid reader. While perusing an old book on herbal medicine, I read how the little old ladies of Mexico made and used a cannabis/tequila rub on their arthritic hands. Then I met Joey, an epileptic musician. He told me another interesting fact- when he had pot he could cut his medication in half! On a camping trip years later, I smelled an unmistakable odor. Following my nose, I was totally shocked to find a grandmotherly lady in her 70s puffing away on a delicate oriental pipe. “Parkinson’s. And the pot’s way cheaper than the pills!” Her nephew kept her well supplied, she said. We had a nice chat about various medical uses of cannabis.

Epilepsy, Parkinson’s, arthritis, and my migraines! What else was it good for? Yet every news article on cannabis that I saw, claimed one new horror after another. Men grew breasts and were impotent. Women became sterile or miscarried. It made you crazy and murderous. Made you lazy and do nothing. It caused cancer and heart attacks…What I had learned on my own and from others and what I was being told in the press were so different!

What was the truth? I began researching. I printed the first studies up and kept them in a notebook, just as a personal reference. The notebook quickly filled. I started a Word file of the URLs and on July 30 2007, I posted it. It continues to grow.

Here’s some of what I have found. All I’ve done is copy the URLs, then put them all in some semblance order for everyone to use as a reference. Please feel free to share this list with anyone who could benefit from it.

Cannabinoids are important chemotaxonomic markers unique toCannabis. Previous studies show that a plant’s dry-weight ratioof {Delta}9-tetrahydrocannabinol (THC) to cannabidiol (CBD) can beassigned to one of three chemotypes and that alleles BD andBT encode alloenzymes that catalyze the conversion of cannabigerolto CBD and THC, respectively. In the present study, the frequenciesof BD and BT in sample populations of 157 Cannabis accessionswere determined from CBD and THC banding patterns, visualizedby starch gel electrophoresis. Gas chromatography was used toquantify cannabinoid levels in 96 of the same accessions. Thedata were interpreted with respect to previous analyses of geneticand morphological variation in the same germplasm collection.Two biotypes (infraspecific taxa of unassigned rank) of C. sativaand four biotypes of C. indica were recognized. Mean THC levelsand the frequency of BT were significantly higher in C. indicathan C. sativa. The proportion of high THC/CBD chemotype plantsin most accessions assigned to C. sativa was <25% and inmost accessions assigned to C. indica was >25%. Plants withrelatively high levels of tetrahydrocannabivarin (THCV) and/orcannabidivarin (CBDV) were common only in C. indica. This studysupports a two-species concept of Cannabis.

Gas chromatography and liquid chromatography differ in one way that is important to the analysis of cannabinoids. Gas chromatography uses heat and liquid chromatography does not. Heat allows the sample of marijuana to go through a chemical process called decarboxylation, which is a necessary step to determine the exact amount of CBD and THC that can be absorbed into the body. The body will not absorb marijuana that has not been decarboxylated, which is why prior to a patients use marijuana is always exposed to heat. Marijuana is decarboxylated either through smoking, vaporizing, or cooking butter.

Analyzing medical marijuana without decarboxylation, like through liquid chromatography, allows only for the measurement of the maximum THC and CBD possible in a sample. This produces high numbers that are not useful to patients because they will need to decarboxylze marijuana in order to feel its effects.

This is why Gas Chromatography has been the meathod of choice for the quantification of cannabinoids by both the United Nations Office on Drugs and Crime (UNODC) and the National Institue on Drug Abuse (NIDA).

Abstract:

A GC–MS method was performed to determine the total 9-THC content in both drug– and fiber-type cannabis seeds. Drug-type seeds were found to contain much higher levels of 9-THC (35.6–124 g/g) than fiber (hemp) seeds (0–12 g/g). The majority of 9-THC was found to be located on the surface of the seeds. Approximately 90% of the total 9-THC was removed by a simple, quick wash with chloroform. Washed drug-type seeds contained less than 10 g/g. Separation of the seeds into the kernel and testa showed that the bulk of 9-THC is located in the testa, mainly on the outside. The kernels of drug- and fiber-type cannabis seeds contained less than 2 and 0.5 g 9-THC/g seeds, respectively. Fluctuations in the 9-THC content of different replicates of the same type of seeds could be the result of the degree of contamination on the outside of the seeds.

1.1 Background

Cannabis products are the most widely trafficked drugs worldwide, accounting for 65 per cent of all global seizure cases (1.65 million cases) in 2006. 5,200 metric tons of herb and 1,000 metric tons of resin were seized in 2006. Practically all countries in the world are affected by cannabis trafficking. Similarly, cannabis also remains the most widely used drug worldwide, with an estimated 166 million people having used cannabis in 2006, equivalent to some 4 percent of the global population aged 15-64.

At the same time, especially since the end of the last century, production methods have become increasingly sophisticated, resulting in the availability in illicit markets of a wide range of cannabis products with widely varying levels of the main psychoactive ingredient, delta-9-tetrahydrocannabinol (THC). Most recently, there has lso been a renewed debate about increasing THC content (frequently referred to as “potency”) in illicit cannabis products. All of this requires analytical data which are comparable between laboratories and over time.

However, most countries do not require by law the detailed analysis of the THC content of the different products, and where such analyses are carried out, they use a variety of approaches and experimental designs, reducing the comparability of results. For example, the conversion of natural constituents, such as tetrahydrocannabinolic acid (THCA), by both smoking and under certain analytical conditions into THC, and how this should be reflected in the analytical report, are issues which are not yet standardized worldwide.

On the technological side, the analysis of cannabis products is further complicated by the relatively restricted availability of pure or well defined reference material of THC and other cannabinoids.* The present manual is an updated and significantly revised version of the manual on “Recommended methods for testing cannabis” (ST/NAR/8), which was published in 1987. It has been prepared taking into account both developments in analytical technology and advances in the science of cannabis, and with a view to providing the analytical basis for an objective discussion about changes in THC content over time, and differences between regions and products.

Patent /Trademark Cannabis / Medical Marijuana Strains/ Varieties Products and Services

After over 150 applications for trademarks under the U.S. Patent Office’s proposed “Processed plant matter for medicinal purposes, namely medical marijuanacategory, the agency has closed the category due to the, shall we say, ridiculous response. The official reason is because the category was redundant with others, such as business names and manufacturing processes.

Many of the applications for trademarks fall under that former category, such as Colorado’s ReLeaf Center and Total Health Concepts, while some were for specific service names, like Nug Source (an ad company). Foods incorporating marijuana were also a big item for trademark patent, including the now infamous “Dr. Canna Cola.”

The well-known Meyer Keegan from Colorado Springs might take the award for largest number of trademarks and patents applied for by a single individual in the now-defunct medical marijuana category. Keegan applied for trademarks on many terms, such as “ganja,” “Maui Waui,” “Bubblegum,” “Island Sweet Skunk,” and other common strains and smoker terms.

Although the category has been stricken, businesses in the medical marijuana and related fields can still apply for patents and trademarks on their business names, proprietary product names, brands, etc. just as any other business does. The removal of the controversial “medical marijuanacategory from the Trademark and Patent Office does not preclude these businesses from operating as would any other business in any other industry.

The so-far undecided patents that were filed with the TPO before closure of the category are still being vetted. Most agree that while it was a nice try, most of Keegan’s attempts at claiming ownership of common names will probably fail. So you won’t have to change your business or product names to avoid a lawsuit for using words like “ganja.”
